Sampling Methods

A amostra corresponde a folhas de plantas de milho (Zea mays) sintomáticas coletadas em plantações comerciais do estado do Paraná-BR

Study Extent fitopatologia

Method step description:

  1. As folhas foram maceradas com nitrogênio líquido, a extração do RNA total foi feito utilizando-se o reagente Trizol (conforme especificações do fabricante); realizou-se, então, a RT-PCR com primers específicos para a capa proteica do vírus e posteriormente a clonagem utilizando o kit Topo Ta (Invitrogen, conforme recomendações do fabricante) e posteriormente as amostras foram enviadas para um laboratório terceirizado e sequenciados. As sequencias foram analisadas com a ajuda de softwares específicos para estes fins.
